LC-MS MS

In this part, a description is going on about fermentation technology and tandem mass spectrometry. This part is going to describe LC-MS-MS instrumentation and their description

Table of Content
  •

Introduction

The full form of LC-MS-MS is “Liquid Chromatography with tandem mass spectrometry”. It is considered to be a powerful technique of analytics that is combined with the separating power of liquid chromatography. The separation has been made with a highly selective and sensitive capability of mass analysis with the triple power of mass spectrometry. There is a difference between LC-MS and LC-MS-MS in his field. LC-MS/MS is HPLC having a mass spectrometry detector whereas LC-MS instruments are an HPLC unit having a mass spectrometry detector. Fermentation technology is an important aspect of this technology and is considered to be a keyword of the present study.

LC-MS/MS

  • It has been seen that LC-MS has been used for “measuring the small molecule components of cellular metabolism”. Liquid chromatography-mass spectrometry is a very preferred tool for this measurement. It can be seen that LC-MS helps to analyze several species.
  • The analysis says that it is a very effective technology for the analysis of food, drug, and also helps in environmental testing. It can be noticed that it helps in the detection of sensitivity capabilities and physical separation. This is a process that helps in both platforms for the analysis of different things.
  • The technique has been used for the identification, separation, and qualification of known and unknown compounds. It is a powerful technique that helps to identify the chemical properties of different molecules.

LC-MS/MS full   form

The “LC-MS/MS” stands for “liquid chromatography with tandem mass spectrometry”.  This is a technique that is used to combine the power of separation in the chromatography with high sensitivity along with the sensitive mass analysis having the capability to triple the mass’s quadrupole into the spectrometry.  In the process, a sample is collected containing the analyst’s interest making them pumped into the stationary phases with the mobile flowing at the high pressure. The interaction of the various chemicals in the stationary phase along with the mobile phase results in the difference in the migration rate through the LC affecting separation in the column. The various varieties of the combination of the phases among the stationary phase with the mobile phases allow the combination to suit the different conditions for the various solutions. The technique’s strength lies with the power of LC to separate at a wider range for different compounds. Along with the wider range of LC, the capability of MS is also important for compounds to be qualified in the higher sensitivity with unique mass.

LC-MS/MS instrumentation

Under the techniques used in “LC-MS/MS,” the whole process is carried out by different components in different units. The LC unit consists of “Pump, auto sampler, column, detector”. The pump makes the effort in delivering the mobile phase with a certain required rate of flow. Next, the auto sampler helps in injecting samples into the solutions to mix the samples in the solution. The columns are made in LC to collect the injected sample in the different columns so that the samples do not mix with each other. The detector is used in analyzing the various samples separated into components.  The “atmospheric pressure ionization (API)” unit is used in the MS to make the pressure of the whole unit. The MS is a system with a vacuum in it there is no presence of air in the system.

Fermentation   technology

Fermentation technology is a process that is involved in the development of various biochemical organisms, reproduction, or even death. This process is used in various forms in the development of pharmaceuticals and the production of foods; it also involves the production of beverages, alcohol in the industry with a large scale of production. The principle of fermentation technology is to make the organism grow in a condition that is sustainable for the organism. In this process, raw materials are provided to the organism to meet their necessary requirements which involve various gasses like carbon, oxygen, or nitrogen. With different glasses, the necessary requirements like the salts, various traces of the elements, and the vitamins needed fermentation technology.

The fermentation technology is generally carried out in containers known as fermentor or bioreactors. Although the design of the fermentor varies with the different kinds of fermentation carried out in different processes so that the final products can be also differ. The various types of fermentors used in the different fermentation processes are “External Recycle airlift, internal Recycle airlift, tubular tower, Nathan and stirred fermentors”. The processes are batch fermentation, Feb -batch fermentation, and continuous fermentation.

Conclusion

The present study deals with a powerful analytical technique that is the LC-MS/MS. The full form of this technique is Liquid Chromatography-Mass Spectrometry. It is an important technique and is used for the separation and identification of different molecules. In this field fermentation technology has also been described and it is considered to be an important technology for the present assignment. It has been seen that different technologies have been used for the presentation of this matter and it has properly described the matter. Therefore the idea can be drawn that LC-MS/MS is a great analytical technique that helps in this matter.

The “Two Dimensional Chromatography” uses the phase equilibrium partitioning principles to the separated proteins, small molecules in complex mixtures based on the varios interactions with the mobile phase and stationary phase.

Thin Layer Chromatography

Thin layer chromatography is a chromatographic technique used to segregate non-volatile varieties. Thin layer chromatography is conducted on a stagnant substrate such as on glass material, plastic material, or aluminium foil, which is blanketed with a narrow coating of adsorbent substance, generally, silica gel is being used, alumina or alumina, or even cellulose.
