Fold purification refers to the number of times a protein preparation is enriched for the protein being purified. If a protein has a concentration of 1nM before purification and a concentration of 10nM after purification the fold purification is 10 fold. The fold purification is usually calculated by measuring the ratio of specific activity before purification and after purification. The specific activity is the ratio of protein content and protein activity of your protein activity.
