In the 3rd example sign of b will be negative otherwise -b/4a will be -ve and we want it to be +ve hence keeping the sign -ve so the overall sign of the equation cant be determined

because that is a condition to keep the equation always positive its explained in the previous video

Sir, during a>0, minima has closed brackets , i.e -D/(4a) is included but in a<0 the Maxima is in open brackets i.e it is not included . Please explain why so?

There is no open or closed brackets..that is (x,y) point and not interval..use ay bracket
