In this  1st lesson I have discussed about structure of DNA .
before understanding structure of DNA we have to deal with some terminology like NUCLEOSIDE,
NUCLEOTIDE,PENTOSE SUGAR, PHOSPHATE GROUP ETC.
because all these are important to study structure of DNA .
After that we have seen little introduction about double helix model of DNA given by Watson and crick.

Structure of DNA.

In this lesson I have discussed about double helix model of DNA. That is proposed by Watson and crick.
DNA is having backbone made of phosphate and sugar .
While base pairs binds to their corresponding base and they are projected inside like steps of the ladder .

In this lesson I have discussed about packaging of 2.2 m long DNA condensed and get packed inside nucleus .
So there are positive charged proteins which are responsible for this packaging and we call them histone proteins .

In this lesson I have discussed about transformation principal given by Griffith.
Griffith conducted experiments on mice and he introduced two strains of bacteria streptococcus pneumoniae inside mice .
And he watched the reaction given by mice separately introducing each strain.
And we will see how he concluded that there must be transfer of some genetic material from bacteria that killed mice in his one of the experiment.

Transformation Principal

In this lesson I have discussed about Hershey and chase experiment conducted using bacteriophage.
Now bacteriophage is virus that infects bacteria.
They infected E coli bacteria with help of bacteriophage.
They used radiolabbled sulphur and phosphorus to label protein and DNA respectively.
After that they found that only radiolabbled phosphorus found in infected bacteria and there was no sulphar in bacteria.
This proves that it was  DNA. Which is transferred from virus to bacteria and it is the actual genetic material.

Hershey and Chase Experiment

In this lesson we have studied about biochemical nature of the substance which transferred from virulent strain of bacteria to non virulent strain and that causes non virulent bacteria to become virulent.
This experiment was conducted by 
Avery , McLeod and McCarthy .
In year 1944.
They used three enzymes RNAase,
Proteases and DNAase .
With the help of these three enzymes they are able to find that actually DNA is the genetic material and it was transferred from virulent strain to non virulent strain of bacteria.

Biochemical Nature of Transforming Principal

In this lesson I have discussed about properties of ideal genetic material.
What properties should be present in any genetic material so that we can call it an ideal genetic material.
So we have discussed few properties which are there in DNA but not in RNA . 
That's why DNA is more stable than RNA .
After that we have seen the difference between DNA and RNA .
We know that RNA is very reactive molecule due to presence of 2' OH group in every nucleotide and that makes it unsuitable for acting as genetic material.

In this lesson I have discussed about machinery required for process of DNA replication.
Also I have discussed about semi conservative model of DNA replication in which one strand will be parenteral strand and one will be newly synthesized.
This semi conservative model was explained by famous experiment of misselson and Stahl.

In this lesson I have discussed about process of replies.
I have discribed the process which requires an enzyme called DNA dependent DNA Polymerase.
This enzyme helps in synthesis of new strands in which one strand will be continuous strand and other will be discontinuous strand.
Also we have seen that whole process start with formation of a replication fork which formed at initial stage.

In this lesson I have given a brief introduction about process of transcription.
Also I have described transcriptional unit which consists of three parts that is 
1 promoter 
2 structural gene 
3 terminator.
Now structural gene have two strands that is template strand and coding strand.
We must know the direction of both strands .
Also the enzyme useful in process of transcription is RNA polymerase.
Which starts from promoter region from where the whole process begins.

Also these both strands are very important for determining sequences of newly formed mRNA .
After that I have introduced you with some terminology like cistrons which are further of two categories
Monocistronic genes  in eukaryotes
And polycistronic genes in prokaryotes.

In this lesson I have discussed about types of RNA .
There are three types of RNA 
1 messanger RNA 
2 transfer RNA 
3 ribosomal RNA
All of these are very crucial to complete the process of central dogma of molecular biology
i,e- DNA >mRNA>proteins 
Also I have discussed individual RNA and their importance in particular process.

In this lesson I have discussed about process of transcription.
There are three steps in process of transcription
1 initiation
2 elongation 
3 termination
Also enzyme DNA dependent RNA polymerase is main enzyme carrying out whole process.
Now Initiation of process occurs as sigma factor binds to polymerase enzyme and than further elongation Occurs and when sufficient length mRNA produced process get terminated by help of rho factor.

In this lesson I have discussed about complexities associated with transcription process.
So after process of transcription the immature mRNA is produced which have to undergo some modifications so that it becomes fully mature and hence it can further undergo process of translation.
So these post transcriptional modification include three processes
1 capping 
2 tailing 
3 splicing.
