Overview

This lessons explains about the Luria Delbruck experiment or the fluctuation test desigined to confirm  which of the two theories of mutation is correct i.e. induced/ adaptation mutation theory or the spontaneous/ random mutation theory

The lesson discusses the different types of mutagens i.e physical mutagen (ionising and non ionising radiation) and chemical mutagens (base analogs and intercalating agents).

This lesson describes Ames test which is used for the detection of potential mutagens,and the limitations associated with Ames test

In this leson we discuss the different types of point mutations i.e. transition mutation ( missense mutation, neutral mutation and silent mutation) and transversion mutation( nonsense mutation). Also we discuss about frameshift mutations.

Here we discuss about photoreactivation repair/ light repair by enzyme photolyase and repair of alkylating damage.

In the absence of light,the damaged DNA strand can be repaired by excision repair/ dark repair which is of two types i.e. base excision repair and nucleotide excision repair. Here we have discussed the base excision repair mechanisms by DNA gkycosykase and AP endonuclease enzyme

Nucleotide excision repair is another type of excision repair/dark repair which is done by amultisubunit enzyme Uvr ABC endonuclease.

Mismatched DNA bases becomes fixed as mutations in the enxt replication cycle. Thus they are repaired after the DNA replication has occured by mismatch repair mechanism.

Mismatch repair

Various mutagens can damage one or mkre DNA bases such that specific base pairing cannot occur. Such damage is serious for DNA replication as replication enzymes have difficulty at bases in template strand that cannot specify a complementary base pair on the new strand.
Thus gaps are left after the replication fork has passed by. The SOS response operates to allow repair leading to cel survival. Two important genes are: lex A and rec A
